Is it possible to save loaded custom panels or do I have to load the panel each time I open TVPaint?

Oh ! That's because you have a network installer : go to Edit > Preferences > Startup > and choose Startup configuration = Remember last used configuration.
Any change you will bring to the interface will be saved (by default, network version are reset when you quit TVPaint)
